Abstract:
Objective
To investigate the effect of electroacupuncture(EA)on nerve regeneration and motor function recovery of rats after cerebral infarction,and to explore its possible mechanism.
Methods
Eighty healthy male SD rats,2~3 months old,weighing 100~150 g,were included in this study.The rat model of stroke-prone renovascular hypertensive(RHRSP)was established.After 10 weeks of preparating RHRSP model,rats with averaging blood pressure≥180 mmHg,weighing 400~450 g,no neurological deficit symptoms were replication middle cerebral artery occlusion(MCAO)model using electrocoagulation.Digital randomization method was used to divided the rats into sham operation(SM)group,cerebral infarction(CI)group,and electroacupuncture(EA)group,24 rats for each group.The RHRSP rats in CI group and EA group were performed the left temporal craniotomy,and MCAO model was established.According to Garcia test,neurobehavioral score was assessed in 1 day,3 days,5 days and 7 days after cerebral infaction.The sizes of cerebral infarction and the number of neuronal and glial cells were compared between groups by HE staining.The positive cell numbers of Nestin,glial fibrillary acidic protein(GFAP),growth associated protein-43(GAP-43)were observed through immunohistochemistry staining between groups.Single factor variance analysis was used to compare the above data between groups.LSD-t test was used to analyze the data between two group.
Results
A total of 75 SHRSP rats were successfully established.After 10 weeks 48 MCAO model rats were successfully established.The neurological function score of Garciain was significantly higher in EA group than that in CI group,and the difference between the two groups was statistically significant in 3 days,5 days and 7 days groups(t=6.14,2.98 and 2.93,P<0.05).Under microscope,the sizes of infarct volume were significantly reduced in EA groups of 7 days,14 days,21 days than those of CI groups(t=3.50,3.92 and 3.45,P<0.05),but no significant difference was found in 28-day group.The positive cell count of Nestin was significantly increased in different time points of EA group than those of CI group(t=3.85,5.61,2.84 and 3.45,P<0.05).The positive cells of GAP-43 was significantly increased at 7 day and 14 days of EA group compared with those of CI group(t=3.72 and 2.64,P<0.05).The positive cells of GFAP was significantly lower in EA group than those of CI group at 7 days,21 days and 28 days(t=3.02,2.83,3.06,P<0.05),especially the 7-day group.
Conclusion
Electroacupuncture can regulate the proliferation of glial cells and neural stem cell potential,which can improve neurogenesis and promote the recovery of nerve function.
