目的

探讨加味温胆汤对抑郁模型大鼠海马神经肽生长抑素(SS)、P物质(SP),神经肽Y(NPY)水平的影响。

方法

将清洁级SD雄性大鼠40只随机分为空白组、模型组、西药组和中药组,每组10只,应用孤养结合慢性不可预见性应激方法造模,采用放射免疫法检测各组大鼠海马神经肽SS、SP、NPY、CRH含量。所有数据采用统计软件SPSS 11.5处理分析,检测结果以均数±标准差表示(±s),多组间差异显著性采用单因素方差分析(one-way ANOVA),用Student-New man-Keuls法进行组间的两两比较,以P﹤0.05为差异有统计学意义。

结果

正常组SS、NPY分别为(4.44±0.87)pg/mg、(28.24±1.85)pg/mg,模型组大鼠海马SS、NPY分别为(3.54±0.33)pg/mg、(21.89±2.56)pg/mg,与正常组比较含量下降(均P﹤0.05),中西药组SS含量为(4.75±1.08)pg/mg,与正常组比较明显上升(P﹤0.05),西药组NPY为(27.57±8.28)pg/mg,较模型组升高(P﹤0.05);正常组SP、CRH含量分别为(1.50±0.42)pg/mg、(0.57±0.24)ng/mg,模型组SP、CRH含量分别为(3.82±1.02)pg/mg、(0.67±0.11)ng/mg,与正常组比较明显上升(均P﹤0.05),而中西药组分别为(2.72±0.56)pg/mg和(0.45±0.16)ng/mg,与模型组比SP、CRH含量下降(P﹤0.05)。

结论

海马神经肽SS、SP、NPY、CRH水平变化与抑郁发病密切相关,降低海马组织SP、CRH水平、升高SS水平可能是加味温胆汤有效抗抑郁作用机制之一。

Objective

To explore the influence of Jia Wei Wen Dan Tang on hippocampal neuropeptide levels of somatostatin(SS),substance p(SP)and neuropeptide Y(NPY)levels in depression rat model.

Methods

Forty male SD rats were randomly divided into blank group,model group,Western medicine group and Chinese medicine group,10 for each group.Chronic unpredictable stress method was applied to build depression rat model.Contents of SS,SP,NPY and CRH were detected by radioimmunoassay in four groups.Data were analysed using statistical software SPSS 11.5 processing analysis were expressed in mean standard deviation(±s).Significant difference between groups was analysed using single factor analysis of variance(one-way ANOVA),and was compared with a Student-New man-Keuls method.P-values less than or equal to 0.05 were considered statistically significance.

Results

The levels of SS and NPY,were(4.44±0.87)pg/mg and(28.24±1.85)pg/mg in normal group.The levels of SS and NPY were(3.54±0.33)pg/mg,(21.89±2.56)pg/mg in model group,which were significantly deceresed compared with those of normal group(P﹤0.05).The level of SS was(4.75±1.08)pg/mg in Western and Chinese medicine group,which was significantly higher than that of normal group(P﹤0.05).The level of NPY was(27.57±8.28)pg/mg in Western medicine group,which was significantly higher than that of model group(P﹤0.05).The SP,CRH levels were significantly increased in model group(3.82±1.02)ng/mg and(0.67±0.11)ng/mg compared with those of normal group[(1.50±0.42)ng/mg and(0.57±0.24)ng/mg,P﹤0.05].The SP and CRH levels were significantly decreased in Chinese and Western medicine group[(2.72±0.56)ng/mg and(0.45±0.16)ng/mg]than those of model group(P﹤0.05).

Conclusion

Changes of neuropeptide levels of SS,SP,NPY and CRH in hippocampus are closely related with the onset of depression.The effective antidepressant mechanisms of Jia Wei Wen Dan Tang are partly by decreasing levels of SP and CRH and increasing levels of SS in hippocampus.
