目的

观察针刺腧穴“降脂方”对高脂血症大鼠羟甲基戊二酰辅酶还原酶(HMG-CoA)、低密度脂蛋白受体(LDL-R)、胆固醇7α-羟化酶(CYP7A1)的影响,进而探讨针刺“降脂方”逆转高脂血症的初步机制。

方法

44只大鼠用随机数表法随机分为正常组17只、模型组27只,分别给予普通饲料、高脂饲料喂养。3周后各组随机选7只经腹主动脉采血,观察总胆固醇(TC);三酸甘油(TG);低密度脂蛋白-胆固醇(LDL-C)及高密度脂蛋白胆固醇(HDL-C)含量变化。再将模型组随机数表法随机分为对照组和降脂方组进行干预。4周后观察各组大鼠TC、TG、LDL-C、HDL-C的含量、用Real-time PCR方法进行测定肝脏中的HMG-CoA、LDL-R和CYP7A1 mRNA表达。采用SPSS13.0统计软件数据分析,数据资料为计量资料,且呈正态分布,以均数±标准差()表示,两组间比较采用成组设计资料的t检验,3组间比较采用单因素方差分析,以P<0.05为差异有统计学意义。

结果

喂养3周后,与正常组比较,大鼠血清TC、TG、LDL-C升高,HDL-C降低(t=4.09、3.32、5.22、14.57,P<0.01或P<0.05),模型组为高血脂状态。分组后模型组与正常组比较:肝脏中HMG-CoA表达显著高于正常组,LDL-R、CYP7A1水平显著低于正常组(t=54.52、11.78、6.02,均P<0.01);干预4周后,腧穴“降脂方”组与模型组比较:大鼠血清TC、TG及LDL-C降低,HDL-C升高(t=2.67、2.77、2.76、3.74,均P<0.05);肝脏中HMG-CoA表达降低显著,LDL-R、CYP7A1水平明显升高(t=34.99、4.18、12.95,均P<0.01)。

结论

腧穴“降脂方”对大鼠高脂血症具有较好的防治作用,其作用机制与降低血脂、逆转肝脏组织中与脂代谢相关酶的表达有关。

Objective

To observe the effect of electric acupuncture acupoint recipes“lipidlowering”on hydroxymethylglutaryl-coenzyme A (HMG-CoA), low density lipoprotein receptor (LDL-R)and cholesterol 7 alpha hydroxylase (CYP7A1) in fatty liver rat model with hyperlipidemia, and the mechanisms thereof.

Methods

Forty-four SD rats were randomly divided into two groups: control group(n=17) and model group (n=27). The rats of control group were fed standard diet and rats in model group were given high fat diet. Blood samples were collected from 7 rats of each group after 3-week treatment.Changes of total cholesterol (TC), triglyceride (TG), low density lipoprotein cholesterol (LDL-C) and high density lipoprotein cholesterol (HDL-C) were detected and compared between two groups. The model group was randomly subdivided into control group and lipid-lowering group. Changes of TC, TG, LDL-C and HDL-C were observed after 4-week treatment in four groups. RT-PCR method was used to detect the values of HMG-CoA, LDL-R and CYP7A1 mRNA expression in liver tissue. SPSS13.0 statistical software was used for data analysis. The t-test was used in the comparison between the two groups, and single factor analysis of variance was used for data analyses of three groups. Statistically significance was considered when P<0.05.

Results

After three weeks, compared with control group, serum levels of TC, TG and LDL-C were significantly increased in model group but HDL-C were significantly decreased(t=4.09, 3.32, 5.22 and 14.57,P<0.01 or P<0.05). The model rats were hyperlipidemia status. The expression of HMG-CoA was significantly higher in model group than that of control group(P<0.01), and expression levels of LDL-R and CYP7A1 were significantly lower in model group than those of control group (t=54.52, 11.78 and 6.02,P<0.01). After 4-week acupuncture treatment, the serum levels of TC, TG and LDL-C were significantly decreased, the level of HDL-C was significantly increased, in lipid-lowering group than those of model group(t=2.67, 2.77, 2.76 and 3.74, P<0.05). The expression of HMG-CoA was significantly decreased, and expression levels of LDL-R and CYP7A1 were significantly increased in lipid-lowering group than those of model group (t=34.99, 4.18 and 12.95, P<0.01).

Conclusion

Electric acupuncture acupoint recipes“lipid-lowering” have good effects of prevention and treatment on hyperlipidemia, which are related with lowing blood lipid and reversing the expression of enzymes in liver.
